Conducted by Vanessa Scammell, the orchestra will present The Man From Snowy River in Concert at Arts Centre Melbourne's Hamer Hall from August 17-19.
Bruce Rowland's epic score for the 1982 George Miller film earnt him APRA and AFI awards.
The world premiere event will see the score performed live to a screening of the famous film.
The film's stars Sigrid Thornton and Tom Burlinson will also be on hand to share behind-the-scenes insights, personal experiences and their impressions of the lasting impact of this treasured Australian classic.
Based on Banjo Paterson's famous bush poem, the story follows young Jim Craig (Burlinson) who sets out to become his own man following the death of his father.
After taking a job with horse rancher Harrison (Kirk Douglas), Craig is mistreated by the other cowboys, but wins the heart of the rancher's daughter, Jessica (Thornton).
When an expensive horse goes missing he is unfairly blamed, but sets out to restore his honour and prove he is worthy of Jessica's hand.
Thornton said she was excited for the upcoming event.
"The Man from Snowy River was a transformational experience for me in a number of ways," she said.
"It changed the trajectory of my working life, it enhanced my understanding of filmmaking as storytelling, and it opened my eyes much wider to the magnificence of the Australian landscape.
"I'm very proud to have been part of a film that continues to resonate across the generations, and I'm now especially excited to see the film shown with Bruce Rowland's marvellous soundtrack played live by the Melbourne Symphony Orchestra."
Performances will take place at 7.30pm on August 17 and 18 and 3pm on August 19 and tickets are on sale now.
